摘要
目的探讨达英-35对PCO模型大鼠中卵巢组织cx43表达的影响。方法选用36只6周龄SD雌性有规律的动情周期大鼠完全随机分为正常对照组、模型组和达英-35组(炔雌醇环丙孕酮)组,每组12只。第22天各组大鼠采血,检测血清睾酮、黄体生成素、卵泡生成素水平,采用化学发光方法,符合PCO大鼠模型标准者纳入实验研究,21d后,处死取卵巢,观察卵巢组织学变化;免疫组织化学观察达英-35对PCO模型大鼠中卵巢组织cx43表达。结果①与正常对照组比较,PCOS模型组大鼠卵巢组织颗粒细胞cx43灰度值下降(98.194±6.51),卵巢组织cx43蛋白表达水平下降(0.66±0.15),差异具有显著性(P<0.05)。达英-35治疗组大鼠卵巢组织cx43蛋白表达水平(0.82±0.14),差异无显著性(P>0.05)。②与PCO模型组比较,达英-35治疗组大鼠卵巢组织cx43灰度值上调(143.794±9.81),达英-35治疗组大鼠卵巢组织cx43蛋白表达上调(0.88±0.12)。结论①卵泡颗粒细胞cx43表达异常降低与多囊卵巢综合征的发生有相关性。②达英-35治疗作用可能与提高卵巢组织连接蛋白cx43有关。
Objective To investigate the effect of diane-35 on expression of cx43 inrandomLy divided into three groups: normal group,PCOS model group and Diane-35 group.The rats in model group were administered letrozole with the dosage of 1mg/(kg d) dissolved in 1% solution of carboxymethyl cellulose by irrigate,for 21 days,and rats in the control group were administered with equal dosage and volume of CMC by irrigate for 21 days.Gonadal hormone concentrations in serum were determined by chemoluminescence method.the histological changes in ovarian tissues were observed with HE staining,and the expression of cx43 in ovary tissue was detected by immunohistoehemistry and westernblotting.Result After drug intervention,in comparison with the model group rats,the serum level of T,FSH,LH of the Diane-35 group rats significantly reduce(P0.05),The serum level of sex hormone of normal group and Diane-35 group rats were not changed(P0.05).The ovaries in the model group showed high incidence of subcapsular ovarian cyst and capsular thickening and decreased number of corporalutea comparing to those in the control group.With the observation on and comparison of staining densities,the mRNA transcription and protein depression the cx43 of the model group were significantly lower than those of control group(P0.05).Conclusion The letrozole induced PCOS rat model is an ideal PCOS animal model for studying the pathogenesis of PCOS.The extra higher expression of cx43 in follicle granulosa cells may be significantly correlated with the pathophysiology mechanism of PCOS.The decrease the expression of cx43 in ovary tissue in rats with polycystic ovaries induced by diane-35.[Key words] Polycystic ovarian syndrome;cx43;Western blotting;Diane-35;Rat
