摘要
目的观察等位基因HLA-A*02:09与4种单倍体的关系,以通过筛检相关单倍体缩小模棱两可确认实验的检测范围。方法采用PCR-SSO-Luminex方法检测的HLA-A*02:01和A*02:09的模棱两可样本通过EX-ON4-7补充实验确认HIA-A*02:09阳性样本,并观察是否携带特定单倍体。结果对205例A*02:01/02:09的模棱两可样本进行了确认实验,包含携带4种单倍体的样本58例,3例检出A*02:09;未携带4种单倍体的147例未检出A*02:09。结论 HLA-A*02:01/02:09模棱两可的比例较高,筛检高危单倍体可有效缩小骨髓库入库样本发生A*02:01/02:09这一组模棱两可时的确认实验的检测范围。
Objective To investigate the degree of correlation between the HLA-allele A * 02 : 09 and 4 haplo- types and try to decrease confirmation tests of the ambiguous HLA-alleJe A * 02 : 01 and A * 02 : 09 by screening high risk haplotypes. Methods 1374 random samples from stem cell donors were analyzed using PCR-SSO-luminex and the unidentified samples were confirmed with confirmation test. Results 205 ambiguous HLA-A* 02 : 01 and A* 02:09 were tested in 1 374 samples. 58 of them had 4 haplotypes and 3 were identified as A * 02 : 09, while other 147 samples lacked A* 02 : 09. Conclusion There was high frequency of ambiguous HLA allele A* 02 :01 and A* 02 :09. Screening high risk haplotypes can effectively decrease the confirmation test of HLA-A * 02 : 01 and A * 02 : 09 ambiguities.
