丝裂霉素C局部应用对SD大鼠坐骨神经组织形态学及超微结构的影响 被引量:1

Effect of topical application of mitomycin C on morphological changes and ultrastructure of rat sciatic nerve

摘要 目的观察局部应用丝裂霉素C(MMC)对大鼠坐骨神经组织形态学及超微结构的影响。方法 SD大鼠30只随机均分成6组。麻醉后暴露右侧坐骨神经,充分游离,分别以棉片浸透生理盐水(NS,B组)及不同浓度的MMC溶液0.1、0.3、0.5、0.7mg/ml(C、D、E、F组),包绕两侧坐骨神经5min;对照(A组)组不做任何处理。分别于敷后即刻、24、48、72h用肉眼观察、光镜及电镜结果评估坐骨神经的形态学及超微结构改变。结果与对照组比较,实验组在用药72h内肉眼观察坐骨神经大体外观、组织形态学观察神经外膜、神经有髓纤维数量及超微结构观察髓鞘厚度、轴突直径差异均无统计学意义。结论局部应用不同浓度MMC(浓度不大于0.7mg/ml)对SD大鼠坐骨神经形态学及超微结构无明显影响。 Objective To investigate the effect of topical application of mitomycin C(MMC) on morphological changes and ultrastructure of rat sciatic nerve. Methods Thirty SD rats were equally randomized into six groups. After anesthesia, the sciatic nerves were exposed fully under sterile and covered by a cotton sheet with normal saline(group B) or with different concentrations of MMC 0. 1, 0. 3,0.5 and 0. 7 mg/ml(groups of C,D,E and F) for 5 min,while no treatment was applied in group A as a sham control. Morphological changes of the sciatic nerve were evaluated by naked eye observation, light microscopy and electron microscopy at immediate time and on the 1st , 2nd, 3rd day after treating. Results The morphology and ultrastructure of the sciatic nerve in the treating groups of B,C,D,E and F were all similar to those in group A(P〉0. 05). Conclusion There is no significant influence of topical application of MMC with a concentration not higher than 0. 7 mg/ml on morphological changes of the sciatic nerve in SD rats.
