摘要
以B2、B5及B8 3个转ChIFN-α基因百脉根株系为材料,通过GUS基因组织化学染色、ChIFN-α基因PCR扩增和基因测序检测,探讨转基因植物中外源基因在无性继代培养条件下的稳定性。结果表明:愈伤组织继代培养12代后,整合于百脉根基因组中的外源ChIFN-α基因序列仍保持稳定,未出现序列变化。
Transgene stability is pivotal for the utilization of transgenic plants. The genetic stability of ChlFN- a gene from different subculture cycles of transgenic Lotus corrticulata callus B2, B5 and B8 were investigated using GUS staining, PCR and sequeneing methods in the current work. The result showed that compared with the original sequence of the target gene, no difference in ChlFN-α gene was observed from the callus subcul- tured as long as 12 cycles.
