GATA-4在不同能量水平下绵羊睾丸中的表达与定位分析

摘要 为了研究GATA-4在不同能量水平下绵羊睾丸中的表达特点及定位情况,提取睾丸组织总RNA,应用Real-Time PCR方法检测睾丸中GATA-4mRNA的表达规律,利用免疫组化技术对睾丸中GATA-4进行定位分析。结果表明,GATA-4mRNA在睾丸中的表达水平由低到高依次为:35%VFI组﹤65%VFI组﹤自由采食组;免疫组化结果显示睾丸Sertoli细胞、Leydig细胞、生精细胞均有阳性产物,阳性强度分别为:35%VFI组﹤65%VFI组﹤自由采食组。GATA-4基因在不同能量水平下具有表达差异性,对绵羊的繁殖性能具有重要的调控作用。 To study the expression of GATA 4 and localization of its protein in the ram testes feeding diets with different energy levels, total RNA was isolated from testes, and mRNA expression was detected by real-time fluorescent quanti- tative PCR. I.ocalization of GATA-4 in testes was examined by immunohistochemistry. The quantitative real-time PCR results showed that the rank of GATA-4 mRNA expression was 35% VFI group ~ 65o/00 VFI group "~ Voluntary feed intake (VFI). The immunohistochemistry showed positive expressions of GA TA-4 in Sertoli, Leydig and spermatogen ic cells of ovine testes, and the rank of positive intensity was 35~ VFI group ~ 65~ VFI group ~ Voluntary feed in- take (VFI). GATA-4 appeared to he differentially expressed at different energy levels, which plays an important regu- lative role in the reproductive performance. Furthermore, the function mechanism of GATA-4 needs to be further re- searched.
