寒、热方剂对胃溃疡模型大鼠炎症因子含量的影响

Impacts of Cold or Heat Prescriptions on Inflammatory Factor Contents for the Model Rats with Gastric Ulcer

目的探讨醋酸涂抹型胃溃疡寒、热证模型的形成机制及寒、热方剂对该模型的作用机理及其方证相应的客观规律。方法采用寒、热因素结合醋酸涂抹法建立大鼠胃溃疡寒、热证模型,采用放射免疫法检测受试大鼠血清中IL-8、TNF-α的含量。结果醋酸涂抹型胃溃疡热证模型中IL-8、TNF-α含量明显高于空白组(P<0.01);且热醋组、热假组含量显著高于寒醋组、寒假组(P<0.05或P<0.01);病证结合模型较单纯证或病模型其含量升高明显。寒性方剂对醋酸胃溃疡热证模型大鼠IL-8、TNF-α含量的升高具有降低作用;而热性方剂对该模型无作用。结论通过"方证相应"理论验证了"病证结合"胃溃疡寒、热证模型的可行性,大黄黄连泻心汤对热性胃溃疡的治疗作用,可通过改变大鼠血清中IL-8、TNF-α的含量而实现。

Objective To explore the mechanism of gastric ulceration induced by acetic acid smear test combined with cold or heat pattern/syndrome and the function mechanism of cold or heat prescriptions on the models as well as the objective laws corresponding to the prescriptions and patterns/syndromes of traditional Chinese medicine. Methods The cold or heat factors in combination with acetic acid smear test were used to establish the models of gastric ulcer with cold or heat pattern/syndrome in rats. The radioimmunoassay（RIA） was adopted to detect the contents of IL - 8 and TNF - c~ in serum for the rats in the trial. Results For the heat - pattern/syndrome model of gastric ulcer induced by acetic acid smear test, the contents of IL -8 and TNF -oL were higher apparently than those in the blank group（P 〈 0.01 ）. The contents in the heat acetic acid group and the pseudo - heat group were higher significantly than those in the cold acetic acid group and the pseudo - cold group（ P 〈 0.05 or P 〈 0.01 ）. The contents were increased apparently for the disease- pattern/syndrome correspondence model as compared with the simple pattern/syndrome model or disease model group. The prescriptions of cold nature decreased the contents of IL - 8 and TNF - ct for the model rats with gastric ulcer induced by acetic acid combined with heat pattern/syndrome. But the prescriptions of heat nature had not impacts on this model. Conclusion The theory of the prescription - pattern/syndroine correspondence verifies the feasibility of the coordination of disease and pattern/syndrome in gastric ulcer models with cold or heat pattern/syndrome. Dahuang huanglian xiexin decoction brings the effect on gastric ulcer of heat type probably through altering the contents of IL - 8 and TNF - ct in rat serum.
