摘要
目的建立石榴皮提取物中鞣花酸含量的HPLC测定方法。方法色谱条件:色谱柱为Agilent TC-C18(2)(4.6mm×250mm,5μm);流动相为乙酸乙酯∶磷酸(0.01mol/L)∶磷酸二氢钾(0.01mol/L)∶甲醇(0.05∶20∶20∶60);流速1mL/min;检测波长为254nm;柱温25℃。结果鞣花酸在5.36~171.40μg/mL范围内与峰面积呈良好的线性关系,平均回收率为97.82%,RSD为1.41%(n=9)。结论 HPLC法简便,准确,重复性好,可用于石榴皮提取物中鞣花酸的含量测定。
Objective To establish an high pressure liquid chromatography(HPLC) method for the determination of ellagic acid in the extract of pomegranate peel.Methods The determination was carried out on an Agilent TC-C18(2)column(4.6 mm×250 mm,5 μm) with mobile phase consisted of ethyl acetate,0.01 mol/L KH2PO4-H3PO4,methanol(0.05∶20∶20∶60) at a flow rate of 1.0 mL/min.The detection wave length was 254 nm and the column temperature was at 25℃.Results The linear range of ellagic acid was 5.36~171.40 μg/mL.The average recovery was 97.82% and RSD was 1.41%(n=9).Conclusion The method is simple,accurate and reproducible for determination of ellagic acid in the extract of pomegranate peel.
