摘要
目的探讨香青兰颗粒对肾性高血压大鼠血压、血流动力学的影响及其降压机制,为传统药物新用提供实验依据。方法采用左肾动脉狭窄法建立两肾一夹高血压大鼠模型;设假手术组、模型组、香青兰组、卡托普利组,每组8只;每天灌胃给药1次。无创尾套法测量大鼠尾动脉收缩压;麻醉状态下分离大鼠颈总动脉,利用左心室插管测血流动力学指标;ELISA法测定血清、心肌组织和肾组织中一氧化氮(NO)水平与血浆、心肌组织和肾组织中内皮素(ET)水平。结果与模型组相比,香青兰组肾性高血压大鼠尾动脉收缩压显著降低;颈动脉收缩压(SBP)、舒张压(DBP)、左室收缩压(LVSP)、左室舒张末期压(LVEDP)降低,室内压最大上升速率(+dp/dtmax)、室内压最大下降速率(-dp/dtmin)和室内压对数值最大变化速率(+dp/dtmax)/LVSP和(-dp/dtmin)/LVSP升高;NO水平升高,ET水平降低。结论香青兰颗粒能降低肾性高血压大鼠血压并对血流动力学产生一定影响,其机制可能是通过调节NO和ET水平改善内皮功能。
Objective To observe the effects of Dracocephalum Moldavica(DM) granule on blood pressure in renovascular hypertensive rats(RHR) and explore the mechanisms.Methods Two-kidney-one-clip RHRs were obtained by narrowing left kidney arteries.4 groups were divided as control,model,DM and captopril groups,each group had 8 rats.The drugs were given by intragastric administration at 1 time per day for 6 weeks.Systolic blood pressure(SBP) and diastolic blood pressure(DBP) were measured by tail cuff approach.In anesthetic conditions,split common carotid artery and utilize left ventricular incubation was used to measure hemodynamic parameters.Levels of NO in serum,heart and kidney and endothelin(ET) in plasma,heart and kidney respectively by enzyme linked immunosorbent assay(ELISA) approach.Results Compared with model group,DM granule could reduce RHR blood pressure including both SBP and DBP as well as reduce both the LVSP and the LVEDP while raise +dp/dtmax,-dp/dtmin,(+dp/dtmax)/LVSP and(-dp/dtmin)/LVSP.In addition,DM granule increased the concentration of NO while decreased the content of ET.Conclusion DM granule can reduce RHR blood pressure and improve the hemodynamics,which may be associated with adjusting the levels of NO and ET to develop the endothelial function.
