摘要 目的考察AB-8大孔树脂分离纯化柿叶总黄酮的工艺条件及其参数。方法以总黄酮回收率为指标,采用单因素试验及正交试验,对大孔树脂分离纯化柿叶总黄酮的工艺进行优化,并对比分离前后总黄酮的纯度。结果优化的工艺条件,上样量为150 mg,上样质量浓度为3.75 g/L,上样药液pH为5~6,乙醇洗脱浓度为80%,乙醇洗脱速率为1 BV/h。平均总黄酮回收率达到89.54%,分离后纯度提高到43.38%。结论用该工艺条件对柿叶总黄酮进行分离纯化,成本较低,分离效果好,可为工业生产提供参考。 Objective To study the processing parameters of the purification of flavonoids from persimmon leaf by macroporous resin. Methods The purification process of flavonoids from persimmon leaf were optimized by single- factor method and orthogonal experiment with reclaim ratio of flavonoids as index. And the purity of flavonoids before and after separation were determinated and compared. Results The sample volume was 150 mg; the concentration of sample was 3.75 g/L; the pH of sample liquid was 5- 6; the ethanol elution concentration was 80% and the speed was 1 BV/h. The average recovery of total flavones was 89.54% and product purity was 43.38% after separation. Conclusion The process condition for the separation and purification of flavonoids from persimmon leaf has low cost and good separation. It can be referenced for industrial production.
