摘要
目的 :建立人血浆中万古霉素 HPL C测定法 ,为临床监测血药浓度以调整万古霉素剂量提供方法学基础。方法 :采用液 -液萃取作提取法 ,并选用去甲万古霉素为内标 ,色谱条件 :YWG- RP1 8分析柱 ,流动相为乙腈 - 0 .0 5 m ol/L KH2 PO4(8∶ 92 ) ,紫外 2 36 nm检测。结果 :本法在 1.0 5~ 84μg/ml间线性良好 ,相关系数 r=0 .9998(n=5 )。日内、日间精密度 RSD均 <1% ,绝对回收率均 >70 %。结论 :本方法简便可行 。
AIM: To establish a high performance liquid chromatography (HPLC) assay for the determination of concentration of vancomycin in human plasma. METHODS: The new HPLC procedure involved a liquid liquid extraction pretreatment procedure and a HPLC internal standard, norvancomycin. The sample was separated on C18 analysis column with a mobile phase of mixture of acetonitrile: 0 05 mol/L potassium dihydrogen phosphate (8∶92). The UV detector wavelength was set up 236 nm. RESULTS: A linearity was obtained from 1 05 to 84 μm/ml of vancomycin in plasma with a good correlation ( r= 0 999 8 , n=5 ). The intra and inter day precision were less than 1% and the absolute extraction recoveries were more than 70%. CONCLUSION: The method is rapid, simple and accurate.
