摘要
目的检测RECK及相关MMPs在神经母细胞瘤及肝母细胞瘤细胞中的表达,探讨RECK及相关MMPs在两种肿瘤的转移特性中的不同意义。方法收集1995年1月至2011年2月手术切除并经病理证实为神经母细胞瘤和肝母细胞瘤的蜡块标本73例,神经母细胞瘤36例,肝母细胞瘤37例。男43例,女30例,年龄6个月~8岁,平均为3.6岁。采用第二代通用型二步法监测系统(PV-6000)免疫组织化学方法检测神经母细胞瘤和肝母细胞瘤组织中RECK及相关MMPs的表达情况。结果RECK在肝母细胞瘤组织中的阳性率为43.2%(16/37),高于神经母细胞瘤27.8%(10/36),MMPs在恶性肿瘤的转移特性中有明显意义。神经母细胞瘤及肝母细胞瘤肿瘤组织中RECK和MMP-14、MT1-MMP的表达均呈负相关(P〈0.05,r=-0.418,P〈0.05,r=-0.583)。结论MMPs在神经母细胞瘤中的表达高于肝母细胞瘤;RECK和相关MMPs的表达随肿瘤的转移、浸润呈负相关。
Objective To study the expression of reversion-inducing eysteinerich protein with Kazal motifs (RECK), matrix metalloproteinase (MMPs) in neuroblastoma and neuroblastoma, and investigate the? influence of RECK and MMPs on tumor metastasis. Methods From January 1995 to February 2011, 73 resected tumor specimen including 36 neurohhstoma specimens and 37 neuroblasto- ma specimens were collected and recruited in this study. PV 6000 immunohistochemical study was performed to investigate the expressions of RECK and MMPs in neuroblastoma and hepatoblastoma. Results The expression positive rate of RECK in the hepatoblastoma specimens was 43. 2%(16/37), which was higher than? that in neurohlastoma (27. 8%, 10/36). The expression of MMPs in neuro- blastoma and neuroblastoma was correlated with tumor metastasis. The expressions of RECK, MMP- 14 and MT1-MMP in neuroblastoma and neuroblastoma were negatively correlated with tumor metas- tasis (P〈0. 05, r = - 0. 418; P〈(0. 05, r = - 0. 583). Conclusions The expression positive rate of RECK in the hepatoblastoma specimens is higher than that in neuroblastoma. The expressions of RECK, MMP14 and MT1-MMP in neuroblastoma and neuroblastoma are negatively correlated with tumor metastasis and infiltration.
