摘要
目的:观察去势后兔关节假体周围界膜组织中OPG/RANKL的变化情况,探讨性激素骤减对人工关节松动的影响。方法:将30只成年新西兰兔随机分成3组,实验组动物去势、假体及磨屑植入造模;实验对照组不去势而行假体、磨屑植入;空白对照组不做任何处理。在第12周处死动物,取股骨远端及胫骨近端假体外周界膜组织,制成组织匀浆,离心后取上清液,用ELISA法检测定OPG及RANKL的含量。结果:与对照组相比,实验组OPG的表达明显降低,RANKL的表达明显上调(P<0.05),OPG/ANKL变化差异更显著(P<0.01)。结论:假体磨屑植入可促使兔关节假体周围破骨细胞形成活跃,成骨活动受抑制,而因去势导致的性激素骤减可明显加剧这一过程。
Objective:To explore the OPG/RANKL changes in interface tissue around implanted prosthesis in castrated rabbits so as to investigate the influence of sudden hormone decline on prosthesis stabilization.Methods:Thirty New Zealand adult rabbits were randomized into 3 groups:castration with prosthesis implantation group,only prosthesis implantation group,and control group without any intervention.All rabbits were sacrificed at the end of 12 weeks for detection of OPG and RANKL by ELISA in the interface tissue around distal femur and proximal tibia.Results:Compared with control group,OPG expression in castration group was remarkably declined while RANKL elevated(P〈0.05),with more significant change in OPG/RANKL(P〈0.001).Conclusion:Prosthesis implantation can promote osteoclast activity thus suppress bone formation,while sudden hormone decline caused by castration can obviously speed this course.
