摘要
目的探讨CXCL2和HIF-1α在前列腺癌(prostatic cancer,PC)组织中的表达,分析其与前列腺癌临床病理特征的关系。方法对112例前列腺标本[PC(PC组)76例,前列腺上皮内瘤(prostatic intraepithelial neoplasia,PIN;PIN组)15例,良性前列腺增生(benign prostatic hyperplasia,BPH;BPH组)21例]行免疫组化SP法检测CX-CL2和HIF-1α的表达。结果 PC组、PIN组CXCL2、HIF-1α阳性表达率均显著高于BPH组,差异均有统计学意义(均P<0.01);PC组CXCL2、HIF-1α阳性表达率显著高于PIN组,差异有统计学意义(P<0.01)。高分化组CXCL2、HIF-1α阳性表达率显著低于中分化组,差异有统计学意义(P<0.05);高分化组、中分化组CXCL2、HIF-1α阳性表达率均显著低于低分化组,差异有统计学意义(均P<0.01)。A期组、B期组CXCL2阳性表达率均显著低于C期组,差异均有统计学意义(P<0.05或P<0.01),A期组、B期组、C期组CXCL2阳性表达率均显著低于D期组,差异均有统计学意义(均P<0.01)。A期组、B期组、C期组HIF-1α阳性表达率均显著低于D期组,差异均有统计学意义(P<0.05或P<0.01),A期组HIF-1α阳性表达率显著低于B期组、C期组,差异均有统计学意义(P<0.05或P<0.01),B期组HIF-1α阳性表达率显著低于C期组,差异有统计学意义(P<0.05)。PC转移组CXCL2、HIF-1α阳性表达率均显著高于PC无转移组,差异均有统计学意义(均P<0.01)。PC中CXCL2表达与HIF-1α表达呈正相关(r=0.422,P<0.01)。结论 CXCL2、HIF-1α在PC组织中呈高表达。CXCL2、HIF-1α表达与前列腺癌病理分级、临床分期、淋巴结转移等有关。但CXCL2和HIF-1α在PC发生、发展中的作用机制还有待进一步的研究。
Objective To investigate the expression of CXCL2 and HIF-1α in prostate cancer(PC) tissue,and to analyze its relationship to clinical and pathological features of PC.Methods The expression of CXCL2 and HIF-1α was detected by immunohistochemistry in 112 prostate specimens,including 76 PC specimens(PC group),15 prostatic intraepithelial neoplasia(PIN) specimens(PIN group) and 21 benign prostatic hyperplasia(BPH) specimens(BPH group).Results Compared with PIN group,the positive expression rates of CXCL2 and HIF-1α significantly increased in PC group,but obviously decreased in BPH group(P0.01).Compared with mod-erately differentiated tissue,the expression of CXCL2 and HIF-1α significantly increased in poorly differentiated tissue,but obviously decreased in well-differentiated tissue(P0.01).CXCL2 expression in A and B stage groups was lower than that in C stage group(P0.05or P0.01).CXCL2 expression in A,B and C stage groups was lower than that in D stage group(P0.01).HIF-1αexpression in A,B and C stage groups was lower than that in D stage group(P0.05or P0.01).HIF-1α expression in A stage group was lower than that in B and C stage groups(P0.05 or P0.01).HIF-1α expression in B stage group was lower than that in C stage group(P0.05).Furthermore,the expression of CXCL2 and HIF-1α in metastatic PC was significantly higher than that in non-metastatic PC(P0.01).There was a positive correlation between CXCL2expression and HIF-1α expression(r=0.422,P0.01).Conclusion Both CXCL2 and HIF-1α are highly expressed in PC tissue,and the expression is correlated with pathological grade,clinical stage and lymph node metastasis.But,the action mechanism of CXCL2 and HIF-1α in PC occurrence and development remains to be further investigated.
