煤焦沥青诱发大鼠肺癌组织中端粒酶的表达 被引量:1

摘要 目的 :判断端粒酶活性在肺癌中的诊断价值。方法 :将Wistar大鼠随机分为实验组 17只和对照组 8只 ,实验组采用支气管灌注煤焦沥青法诱导大鼠肺癌 ,用改良的TRAP银染法测定大鼠肺癌及正常组织中的端粒酶活性。结果 :实验组诱发出肺癌 8例 ,鳞状上皮化生 1例 ,8例肺癌中有 7例为端粒酶阳性 (87.5 % ) ,鳞状上皮化生的1例也为端粒酶阳性 ,实验组其余 8例与对照组 8例端粒酶均为阴性。有癌组与对照组和无癌组相比差异均有显著性 (P <0 .0 1)。结论 :端粒酶较特异地表达于肺癌组织中 ,有望成为肺癌的诊断指标。 Aim: Judge the value of telomerase activity in the diagnosis of lung cancer. Methods: Wistar rats were random allocated into two groups:17 in the experimental group and 8 in the control one.Use the animal model of rat lung cancer induced by coal tar pitch(CTP) Perfusion, with the modified TRAP(telomerase reaction amplification protocol) silver staining method to examine the telomerase activity of the rat lung cancer and normal tissues. Results: 8 of 17 in the experimental group were induced to lung cancer,also there was 1 speciman with squamous metaplasia, 7of 8 lung cancer were telomerase positive (87.5%) and the one with squamous metaplasia was telomerase positive too. the other 8 of experimental group and the whole 8 of control group were all telomerase negative. the cancer group was significantly different with either non cancer group or control group ( P <0.01).Conclusion: Telomerase activity could be detected in the lung cancer specifically,it could be a hopeful index in the diagnosis of lung cancer.
