摘要
目的测定髁突软骨细胞中细胞因子TNF-α表达水平,探讨应激对颞下颌关节的影响机制。方法选用SD大鼠75只,分为空白对照、电击、应激、药物对照、生理盐水5组,在模拟应激条件下建立颞下颌关节的应激反应动物模型,提取连续刺激10、20、30 d后的各组大鼠的髁突软骨细胞,用ELISA法和RT-PCR测量髁突软骨细胞中TNF-α的含量和TNF-αmRNA的表达水平。结果 ITNF-αmRNA在应激后第1周表达最强,以后逐渐达到正常水平。结论应激能使髁突软骨发生炎性改变,而且这种变化在应激初期表现较为明显。
Objective To detect the expression level of inflammatory cytokines TNF-α in condylar cartilage cell of stressed rat so as to explore the influence mechanism of stress on temporomandibuiar joint. Methods Seventy-five Sprague Dawley (SD) rats were used to establish the animal model of temporomandibular joint responsed to stimulation. The samples of rat condylar cartilage cells of each groups (i e, blank controlled, foot- shocked, emotional stress, anti-anxiety and normal saline, totally 5 groups) at the end of 10, 20 and 30 clays after eontinous stimulation were observed and the levels and mRNA expression of TNF-α were evaluated by ELISA and RT-PCR. Results Obvious mRNA expression of ITNF-α was observed at the 1st week after stimulation, it started to gradually return to normal. Conclusion The stimulation could lead to inflammatory change of condylar cartilage, more significant especially at the initial stage.
