摘要
目的 :探讨基质金属蛋白酶抑制剂 (TIMP 2 )对SGC 790 1胃癌细胞体外侵袭能力的影响。方法 :将人全长TIMP 2基因经脂质体包裹转染PA317细胞系 ,G418筛选 ,感染人SGC 790 1胃癌细胞系 ,经体外细胞增殖、细胞电泳率、软琼脂形成和侵袭小室的实验综合分析其生物学行为改变。结果 :经TIMP 2基因转染的SGC 790 1细胞和MFC细胞和细胞电泳率。软琼脂形成和侵袭能力均明显低于对照组 ,而与对照组相比细胞增殖曲线没有明显的变化。结论 :TIMP 2基因转染可以使肿瘤细胞的表面电荷发生变化 ,使细胞的电泳能力、细胞的移动性和侵袭穿透能力减弱 ,而与细胞的生长周期没有明显影响 ,提示TIMP
Purpose:To study the effect on invasion behavior of gastric carcinoma in vitro of tissue inhibitior of matrix metalloproteinase 2 (TIMP 2) gene. Methods:A recombinant retroviral vector pl MT T2 which contained a flull length TIMP 2 complement DNA was transfered into PA317 cells by the liprofectmin technique. Human gastric cancer cell line SGC 7901 and mouse gastric cancer cell line MFC were used. Cell proliferation,sofa argon and boyden chamber were used to assay the change of invasion ability of transfered cell lines. Results:The electron rate,the ability of invasion of Boyden Chamber were significantly less than in the control groups. The curve ofcells transfered TIMP 2 gene were not significantly different from that of control group.Conclusions:TIMP 2 gene can change the quality of electrical charge on the surface of tumor cells and reverse the malignant phenotype of various type of tumor cells.
