多西紫杉醇对在体和离体人胃癌SGC7901细胞的作用机制的探索 被引量:3

摘要 目的探索多西紫杉醇对在体和离体人胃癌SGC7901细胞的生长抑制作用及其机制。方法将人胃癌细胞种植于培养皿贴壁,用集落形成实验法观察其对体外培养的人胃癌细胞对细胞存活率的影响,光学显微镜和电子显微镜观察形态学改变,并通过流式细胞仪检测细胞周期变化。结果多西紫杉醇可以抑制SGC7901细胞于S期,抑制SGC7901细胞生长增殖,并呈浓度依赖性,差异统计学意义(P<0.05)。结论多西紫杉醇能明显抑制SGC7901细胞增殖。 Objective To explore the sensitization of Doc, etaxel induced human gastric cancer SGC7901 cell growth inhibi- tion in vivo and in vitro. Methods Human gastric cancer cells grown in culture dishes, colony forming experiment assay was used to investigate the cytotoxicity of the human gastric carcinoma cell line in vitro. Light microscopes and electron microscopy were employed to observe the changes in morphology of the cells treated with the drug, flow cytometry was ap- plied to study the effects of the drugs on the cells cycle of the cell lines. Results Docetaxel could arrest the suspended gastric cancer cell to S phase respectively. Docetaxel significantly inhibited growth and proliferation of human gastric can- cer cells in dose-dependent manner (P 〈 0,05). Conclusion Docetaxel may inhibit SGC7901 cell growth.
