布地奈德对哮喘小鼠JAK1及IL-4表达的影响

摘要 目的探讨哮喘小鼠肺组织中JAK1及IL-4的表达及布地奈德对其的干预作用。方法 30只小鼠随机分为对照组(A组)、哮喘组(B组)和布地奈德干预组(C组),C组在激发时给予布地奈德雾化吸入。检测肺泡灌洗液IgE水平、EOS计数和肺组织中IL-4及JAK1表达。结果 B组肺泡灌洗液中EOS计数和IgE水平均较A组高,差异有高度统计学意义(q=11.2,P<0.01和q=6.78,P<0.01)。B组肺组织中IL-4浓度和JAK1表达均较A组高,差异有高度统计学意义(q=9.82,P<0.01和q=7.64,P<0.01)。C组肺泡灌洗液中EOS计数、IgE水平均较B组低,差异有统计学意义(q=4.53,P<0.05和q=3.95,P<0.05)。C组肺组织中IL-4浓度、JAK1表达均较B组低,差异有统计学意义(q=4.13,P<0.05和q=3.56,P<0.05),且JAK1的表达与IL-4的浓度呈正相关(r=0.724,P<0.01)。结论布地奈德可缓解哮喘症状,可能与其抑制肺组织JAK1及IL-4的表达,进而抑制气道炎症的信号传导途径有关。 Objective To investigate the expression of JAK1 and IL-4 in the lung of mice with asthma and the preventive effect of Budesonide on them. Methods 30 mice were randomLy divided into control group (group A), asthma group (group B) and intervention group (group C). Mice in group C inhaled Budesonide when they were challenging. The amount of IgE and EOS in BALF, the expression of JAK1 and IL-4 in lung tissues were detected. Results The amount of IgE and EOS in BALF in group B were higher than in group A, the difference was statistical significant (q = 11.2, P 〈 0.01 and q = 6.78, P 〈 0.01); the expression of JAK1 and IL-4 of the lung tissue in group B were higher than in group A, the difference was statistical significant (q = 9.82, P〈 0.01 and q = 7.64, P〈 0.01). The amount of IgE and EOS in BALF in group C were low- er than in group B, the difference was statistical significant (q = 4.53, P 〈 0.05 and q = 3.95, P 〈 0.05). The expression of JAK1 and IL-4 of the lung tissue in group C were lower than in group C, the difference was statistical significant (q = 4.13, P 〈 0.05 and q = 3.56, P 〈 0.05). JAK1 expression was positively related with IL-4 concentration (r = 0.724, P 〈 0.01). Conclusion Budesonide can relieve the symptoms of asthma, it may be concerned with that it inhibits the signal transduction system of inflammation following inhibits the expression of JAK1 and IL-4.
