PPAR-γ和MMP-9对COPD病人气道杯状细胞增生和MUC5AC表达的影响 被引量:6

EFFECT OF PPAR-γ AND MMP-9 ON HYPERPLASIA OF AIRWAY GOBLET CELLS AND MUC5AC SECRETION IN PATIENTS WITH COPD

摘要 目的探讨气道中过氧化物酶体增殖物激活受体-γ(PPAR-γ)和基质金属蛋白酶-9(MMP-9)对气道杯状细胞增生及黏蛋白5AC(MUC5AC)表达的影响。方法选取男性肺鳞癌手术病人40例,根据肺功能分为COPD组和正常对照组,各20例,取手术切除肺叶的无癌组织浸润的外周支气管组织制备切片,阿辛蓝-过碘酸雪夫(AB-PAS)染色法检测气道上皮杯状细胞百分比,免疫组化法测定气道中PPAR-γ、MMP-9及MUC5AC的含量。结果与对照组比较,COPD组气道上皮杯状细胞数目及气道中PPAR-γ、MMP-9及MUC5AC表达量均明显增高(t=-9.41~-3.85,P<0.01)。COPD组气道中PPAR-γ表达水平与气道上皮杯状细胞百分比、MUC5AC表达水平均呈明显负相关(r=-0.824、-0.865,P<0.01),而MMP-9表达水平与气道上皮杯状细胞百分比、MUC5AC表达水平均呈明显正相关(r=0.875、0.754,P<0.01)。结论COPD气道杯状细胞明显增生、MUC5AC表达增高,而气道PPAR-γ和MMP-9表达异常参与了黏液高分泌的调控。 Objective To investigate the effect of peroxisome proliferators activated receptor gamma (PPAR-γ) and matrix metalloproteinase-9 (MMP-9) on airway goblet cells and MucinSAC (MUCSAC) secretion in airway. Methods Forty patients underwent surgery for squamous cell lung cancer were enrolled in this study. The patients were divided into COPD group and control group according to lung function, with 20 in each group. Cancer-free peripheral bronchial tissue from the resected lung was taken for making pathological sections, the percentage of airway epithelial goblet cell (AEGC) was detected by alcain blue-periodic acid Schiff (AB-PAS) staining and the expressions of mucin, PPAR-γ, MMP-9 and MUC5AC in airway epithelial tissues were measured by immunohistochemistry. Results Compared with the control group, the count of AEGC, the expressions of mucin, PPAR-γ, MMP-9 and MUCSAC were significantly increased in COPD group (t =- 9.41-- 3.85, P〈0.01). The level of PPAR- γ expression was negatively correlated with the percentage of AEGC and MUCSAC expression (r=-0. 824,--0. 865;P〈0.01) in COPD group, while the level of MMP-9 expression was positively correlated with the percentage of AEGC and MUC5AC expression (r= 0. 875,0. 754;P〈 0. 01). Conclusion There is a significant AEGC hyperplasia and high expression of MUCSAC in COPD. The abnormal expressions of PPAR-γ and MMP-9 in airway involve in the regulation of mucus hypersecretion.
