摘要
目的 :探讨聚合酶链反应 (PCR)扩增大片段线粒体DNA方法。方法 :快速碱裂解法提取线粒体DNA ,长时程PCR扩增出 5 430bp线粒体DNA。结果 :扩增出 5 430bp线粒体DNA产物特异性强 ,产量高。 结论 :该方法反应条件简便、稳定 ,也可用于其它大片段的扩增 ,有一定的实用价值。
?Objective:To find the polymerase chain reaction(PCR) method that amplified the long fragment of mitochondrial DNA. Methods:Mitochondrial DNA was isolated by alkaline extraction, the 5 430 bp fragment of mtDNA was amplified by a long PCR.Results:The product of the PCR was specific.Conlusions:The condition of the PCR was simple and stable. The method may also amplify other long DNA fragment. 〔
