摘要
目的 :制备固定化胰蛋白酶 ,用于转肽反应。方法 :以涂敷了壳聚糖的硅胶为复合载体 ,以系统全面反馈优化技术进行优化制备固定化胰蛋白酶。结果 :所制备的固定化胰蛋白酶平均酶活性为 2 2 82U/ g ,酶活性回收在 9%以上。结论 :制备的固定化酶的单位酶活性高 ,带入转肽反应体系的水分少 。
Objective:An immobilized trypsin (ITP) is prepared for transpeptidation. Methods:The silica coated chitosan is used as compound carrier, and the process is optimized by a systemic and all-around feedback optimization technique. Results:The average activity of ITP is as high as 2282U/g, and the recovery rate of activity is 9%. Conclusion:The activity per gramme ITP is very high , and it carries a little water to system of transpeptidation , so it was used for transpeptidation successfully.
