人肿瘤浸润γδΤ细胞的抗肿瘤活性研究

Study on antitumor activity of expanded human infiltrating gamma-delta T lymphocytes

目的研究人直肠癌和结肠癌的肿瘤浸润 γδT淋巴细胞 ( γδTIL s)体外抗肿瘤活性。方法用固相抗体法体外扩增获得γδTIL s,并用 MTT法和 3H- Td R掺入实验体外观察γδTIL s对同种异体和异种肿瘤细胞的细胞毒及增殖反应。结果所获得γδTIL s对所测定的肿瘤细胞显示出明显的细胞毒活性。该杀伤效应可为 IL - 2 ,IL - 4,IL - 12 ,IL - 15 ,TNF-α和 IFN -γ所增强 ,而不受 IL - 10 ,GM- CSF和 TGF-β所影响。某些去增殖活性的肿瘤细胞可诱导静息化的γδTIL s产生显著的增殖反应。结论 γδTIL

Objective To investigate the antitumor activity of selectively expanded human colorectal γδ tumorinfiltrating T lymphocytes(TILs)in vitro.Methods γδTILs were expanded by solid phaseantibody method,whose cytolytic and proliferative activities were detected by MTT method and 3HTdR incorporation.Results Expanded γδTILs demonstrated marked cytotoxicites to allogeneic and isogeneic tumor cell lines.Cytokines including IL2,IL4,IL12,IL15,TNFα and INFγ could promote the cytotoxicities of γδTILs,while IL10,GMCSF and TGFβ had no effect on such killing activities.Rested γδTILs could proliferate in response to some mitomycin Ctreated tumor cells.Conclusions γδTILs could play an important role in antitumor response via their cytotoxicity to tumor cells.
