摘要 目的:观察水通道蛋白1、5(AQPl、5)在人不同病理级别星形细胞瘤组织中的表达差异,探讨星形细胞瘤增殖、生长的分子机制。方法:收集人各个病理级别星形细胞瘤标本55例,以肿瘤周围相对正常脑组织作为对照,采用H—E染色诊断分级,石蜡切片免疫组织化学、免疫印迹分析及逆转录聚合酶链式反应观察AQPl、5及其mRNA的表达变化。结果:与正常脑组织相比,人星形胶质瘤组织中AQPl及其mRNA表达上调,随着星形细胞瘤病理级别的升高,AQPl及其mRNA表达增强,胶质母细胞瘤组织表达最强烈;而AQP5及其mRNA仅在高恶性星形细胞瘤组织中表达增强。结论:AQPl在人星形细胞瘤组织中表达与其病理级别相关,而AQP5仅在人高恶性星形细胞瘤组织中表达增强,提示不同病理级别胶质瘤组织中AQP的表达规律不尽相同。 Objective: To investigate the expression patterns of aquaporin (AQP) 1 and AQP5 in human astrocytic tumors with different pathologic grades, and to explore the molecular mechanism underlying the growth and proliferation of astrocytoma. Methods: Expressions of AQPl and AQP5 in both mRNA and protein level in tumor tissues from 55 patients and normal brain tissues around tumors were detected by means of H-E staining, immunohistochemistry, Western blotting, and reserve transcription polymerase chain reaction. Results: Compared with normal brain tissues, the AQP1 was up-regulated in human astrocytoma, and enhanced with the increasing of pathologic grades. The strongest expression of AQP1 was found in glioblastoma. However, the up-regulated expressions of AQP5 both in mRNA and protein were only found in high-grade astrocytoma. Conclusion: The expression of AQP1 is related to the various pathologic grades of human astrocytoma, while the expression of AQP5 is up-regulated only in high-grade astrocytoma, suggesting that the expression of AQPs is different in tumor tissues of various pathologic grades.
