摘要
单分散、非多孔聚甲基丙烯酸环氧丙酯微球机械强度高,表面的环氧基易于修饰与键合,非常适宜于生物大分子的快速分离、纯化。对其表面环氧基浓度、非特异性吸附性能等进行了表征;并以胰蛋白酶抑制剂为配基,制备了胰蛋白酶抑制剂.交联聚甲基丙烯酸环氧丙酯亲和色谱柱,考察了胰蛋白酶等蛋白质的亲和色谱行为,表明该基质适宜用作亲和色谱载体。提出以添加1%乙腈的方法,可有效地消除此类载体的弱非特异性吸附,使人血清白蛋白的回收率>99%。
A new synthetic support, monosized, non-porous, cross-linked bead CL-poly (glycidyl methacrylate) (CL-PGMA) containing aetive epoxide groups was synthesized and used as the support of affinity chromatogrpaphy. The concentration of the epoxide groups on the surface of CL-PGMA beads was measured and the non-specific adsorpion to protein was charecterized. Trypsin inhibitor was chosen as the ligand. The trypsin was purified 2. 5 times by using the trypsin inhibitor immobilized CL-PGMA affinity column. The chromatographic characterization was shown that the CL-PGMA bead was very suitable for the high performance affinity chromatography. The effective elimination of non-specific adsorption was carried out by adding 1% acetonitrile in mebile phase. The recovery of human serum albumin was more than 99%.
