摘要
目的建造大鼠腹主动脉瘤模型,观察骨桥蛋白(OPN)在实验性腹主动脉瘤中的表达情况,探讨OPN在腹主动脉瘤发病中的意义。方法采用酶灌注法建造大鼠腹主动脉瘤模型,两周后取材并测量大鼠手术后腹主动脉的直径;苏木精-伊红(HE)染色检测主动脉的结构改变及炎性细胞浸润程度;特殊染色检测主动脉中层弹力蛋白变化情况;免疫组织化学技术、蛋白免疫印迹法(Western Blot)法检测动脉组织中OPN的表达量。结果成功建造大鼠腹主动脉瘤模型;腹主动脉瘤组的术后主动脉直径扩张率(220.3%)明显高于对照组(112.3%)和未处理组;HE染色结果显示腹主动脉瘤组中层弹力蛋白明显减少,炎性细胞浸润程度显著增高;免疫组织化学结果显示OPN在腹主动脉瘤组、对照组组及正常组中的表达分别为7.500×1011±9.233×1011、3.073×105±2.581×105、2.037×105±1.866×105(F=6.595,P<0.01);Western Blot法显示OPN在上述3组中的表达分别为2.777×105±2.817×105、4.524×103±1.357×103、2.110×103±1.985×103(F=9.489,P<0.01)。结论 OPN表达量的增加可能加速实验性腹主动脉瘤的发生发展。
Objective To observe the expression of osteopontin(OPN) by making rat model of abdominal aortic aneurysm and explore the significance of OPN in the pathogenesis of abdominal aortic aneurysm. Methods Rat model of abdominal aortic aneurysm was made by perfusing pancreatopeptidase E from hog pancreas into the aorta of rats. Aortas were taken out two weeks later after measuring the diameters of all aortas. HE staining was used to observe the structural change and Elastase staining to observe how the area of elastase altered in the middle tuna of the aorta. Immunohistochemistry and Western Blot were used to detect the expression of OPN. Results Rat model of abdominal aortic aneurysm was successfully made. Compared to saline group and normal group, the infiltration of inflammatory cell was more apparent and the quantity of elastase was significantly lower in experimental group. Immunohistochemistry showed that the expression of OPN in experimental group,saline group and normal group were (7.500×10^11±9.233×10^11)vs(3.073×10^5±2.581×10^5)vs(2.037×10^5±1.866×10^5)(F=6.595,P〈0.01); Western Blot showed the expression of OPN in experimental group, saline group and normal group were (2.777×10^5±2.817×10^5)vs(4.524×10^3±1.357×10^3)vs(2.110×10^3±1.985×10^3)(F=9.489,P〈0.01).Conelusiun The increase of OPN may accelerate the occurrence and development of experimental abdominal aortic aneurysms.
