甘草酸、熊果苷及氢醌对小鼠黑素瘤细胞黑素生成影响的比较研究 被引量:46

摘要 比较观察了 18α 甘草酸双胺盐、熊果苷及氢醌对体外培养B16F10鼠黑素瘤细胞细胞形态、酪氨酸酶活性、黑素含量和细胞增殖率的影响 ,结果发现 3种化合物均显示有抑制黑素生成的作用 ,其中以 5 0 μmol/L熊果苷对酪氨酸酶活性和黑素含量抑制作用最为明显 ,其次是10 0 μmol/L 18α 甘草酸双胺盐 ,但二者并无统计学上的差异 (P >0 0 5 )。氢醌对酪氨酸酶活性的影响呈双向作用 ,即低浓度 ( 0 1~ 0 5 μmol/L)时下调酶活性 ,高浓度 ( 1~ 10 μmol/L)时则能轻度上调。细胞增殖率测定结果显示 18α 甘草酸双胺盐、熊果苷在实验浓度 ( 10~ 5 0 0 0 μmol/L)对细胞增殖率无明显抑制 ,而氢醌浓度超过5 μmol/L时能明显抑制细胞增殖或呈现细胞毒性。细胞形态观察发现 1~ 10 μmol/L氢醌作用2 4h ,细胞胞体变小 ,树状突明显增多且变得细长 ,部分树状突呈串珠状改变。提示 18α 甘草酸双胺盐、熊果苷的皮肤脱色作用机制与氢醌可能不同 ,后者低浓度时以抑制酪酸酶活性为主 ,高浓度时主要是细胞毒 ,且细胞早期的损伤部位可能发生在树状突。 18α Ammonium glycyrrhizinate(18α GL),arbutin and hydroquinone were investigated in vitro for the action on tyrosinase activity, melanin content, and cell proliferation in cultured B16F10 murine melanoma cells The results showed that all of them exhibited an inhibitory effect on tyrosinase activity and melanin production, more marked effect showed by 50μmol/L arbutin, and the 100μmol/L 18α GL, but with no statistical significance between them(P >0 05) Hydroquinone had biphasic effect on tyrosinase activity, at low concentration(0 1~0 5μmol/L) it showed inhibitory effect, and at high concentration(1~10μmol/L) it showed slight up regulation Detecmination of cell proliferation rate showed that 18α GL and arbutin had no marked inhibitory effect on cell proliferation rate at concentretion(10~5000μmol/L),but bydroquinone could markedly inhibit cell prolhiferation rate or showed cytotoxicity when the concentration over 5μmol/L Hydroquinone caused a marked change in cellular morphology, the cell body became small, with many elongate and thin cell processes, some of them were rosary like This effect was observed clearly at final concentration between 1 and 10μmol/L after treating for 24 hours We concluded that the possible mechanisms of hydroquinone bleaching hyperpigmented skin were through its inhibition of tyrosinase activity at lower concentration and its cytotoxicity at higher one
