摘要
目的:探讨单羧酸转运蛋白1(monocarboxylate transporters 1,MCT1)、CD147、CD44 mRNA和蛋白在非小细胞肺癌(non-small cell lung cancer,NSCLC)中表达的相关性及其在NSCLC发生、发展及预后中的意义。方法 :取经病理确诊的56例NSCLC患者癌组织、癌旁组织(距肿瘤边缘>5 cm)和21例肺良性病变组织采用免疫组化和实时荧光定量PCR检测MCT1、CD147、CD44蛋白及mRNA的表达。结果:①在NSCLC组织中MCT1、CD147、CD44表达均明显高于对照组(P<0.05);②MCT1在肺癌细胞膜上的表达水平与CD147、CD44的表达均存在相关性(P<0.05);③MCT1、CD147、CD44的表达与NSCLC患者的性别、年龄、肿瘤大小、TNM分期均无关,但与肿瘤的组织类型(P<0.05)、淋巴转移(P<0.05)显著相关,且MCT1、CD44蛋白和mRNA的表达与肿瘤的分化程度显著相关(P<0.05)。结论:MCT1、CD147、CD44可能参与肿瘤的发生、发展及远处淋巴结转移,可作为检测NSCLC和预后评价的指标。
Objective:To investigate the protein and mRNA expression of monocarboxylate transporters 1 (MCT1),CD147 and CD44 in non-small cell lung cancer(NSCLC), and significance of their clinical pathology. Methods:Fifty-six specimens of the NSCLC and peritumoral tissues and twenty-one control specimens of pulmonary benign diseases were studied. The mRNA and protein expressions of MCT1,CD147 and CD44 in specimens were analyzed by real-time quantitative reverse transcriptase polymerase chain reaction (real-time quantitative RT-PCR) and immunohistochemistry. Results: (1)The positive expression rates of MCT1 ,CD147 and CD44 were significantly higher in NSCLC tissues than those in the peritumoral and control tissues (P 〈 0.05); (2) The MCT1 expression in the cell membrane was correlated with both CD147 and CD44 expressions (P 〈 0.05) ; (3) The expressions of MCT1, CD147 and CD44 in NSCLC were not correlated with age,gender,lesion dimension and TNM stage,but significantly correlated with histological types of tumor and lymph node metastasis (P 〈 0.05). And the mRNA and protein expressions of MCT1 and CD44 in NSCLC was correlated significantly with tumor differentiation degree (P 〈 0.05). Conclusion: MCT1,CD147 and CD44 may participate in the occurrence and development of NSCLC, and may be act as the diagnose and prognosis index of NSCLC.
