一氧化氮对人培养血管内膜增生的抑制作用 被引量:1

Inhibitory effect of NO on neointimal hyperplasia of cultured human saphenous vein

摘要 目的 研究一氧化氮 (NO)对体外培养人正常大隐静脉片内膜增生的影响。方法 取来自同一条正常大隐静脉的 3段长 5mm血管 ,随机分入 3组 ,共用 6条大隐静脉。体外培养 2组大隐静脉片 ,其中 1组加入NO供体亚硝基乙酰青霉胺 (S nitroso N acetylpenicillaminne,SNAP)。免疫组织化学染色后观察内膜增生及细胞增殖。结果 培养静脉片可见明显内膜增生和平滑肌细胞增殖。与对照组相比 ,加入SNAP组培养血管片内膜增生及平滑肌细胞增殖明显减低。对照组内膜厚度为 ( 2 9.0 0± 1 .40 ) μm ,SNAP组为 ( 1 4.0 0± 1 .34) μm ,差异有非常显著性 (P <0 .0 1 )。对照组平滑肌细胞增殖指数为 ( 30 .0 0± 2 .1 3) % ,SNAP组为 ( 1 2 .0 0± 1 .43) % ,差异有非常显著性 (P <0 .0 1 )。 Objective\ To investigate the effect of nitric oxide (NO) on neointimal proliferation in the cultured human saphenous vein. Methods\ Six pairs of segments of human normal saphenous vein were incubated at 37℃ for 14 days in culture medium containing 30% fetal bovine serum. SNAP (a kind of NO donor) was added into one of the paired segments in the concentration of 10 -3 mol/L. Six veins were used for each dose. At the last 4 days, 5 Bromodeoxyuridine (5 BrdU) was incorporated into labelled proliferation cells. The thickness of the neointima was measured by using a computerized image analysis system. The 5 BrdU labelled cells were counted in high power field. The mean percentage of labeled cells was used as the final proliferation index. Results\ Significant reductions in neointimal thickness were seen at 10 -3 mol/L SNAP [control (29.00±1.40) μm vs (14.00±1.34) μm, P <0.01]. The proliferation indices were (30.00±2.13)% in the control group and (12.00±1.43)% in SNAP group respectively ( P < 0.01 ). Conclusion\ NO could inhibit the neointimal proliferation of cultured human saphenous vein.\;
