摘要
目的 研究核转录因子 (NF kB)在心肌缺血预处理中的作用。方法 将 1 8只新西兰白兔随机分为非预处理组、预处理组和预处理加脯氨酸二硫氨基甲酸脂 (ProDTC ,特异性NF kB抑制剂 )组。阻断和开放左冠脉前降支各 5min ,反复 5次 ,造成预处理 ,第 3组在预处理前、中使用ProDTC ( 1 5mg/kg体重 )。电压力探头测定局部心功能 ,氯化三苯四唑啉 (TTC)法计算梗死面积 ,并作组织学检查。结果 预处理对心功能的保护被ProDTC抵消 (P <0 .0 5) ,对梗死面积的减小也被其抵消 ,分别为 ( 8.2± 1 .2 ) %和 ( 38.9± 7.9) % ,差异有非常显著性 (P <0 .0 1 )。ProDTC也消除了预处理组织学保护。
Objective\ To study the role of nuclear factor kappa B (NF kB) in myocardial ischemic preconditioning (IPC). Methods\ Eighteen rabbits were randomly divided into three groups: nonpreconditioned, preconditioned and preconditioned plus proline dithitocarbamate (ProDTC, a specific inhibitor of NF kB activation). Preconditioning was initiated by tightening and releasing the snare around a branch of left coronary artery for 5 min respectively, repeating for 5 times. In the third group, rabbits were administered ProDTC (15 mg/kg) prior to and during IPC. Myocardial segmental shortening was measured with piezoelectric crystals. Infarct size was assessed using TTC method, and histological examination was performed. Results\ The protective effects of IPC on contractile function and the decreased infarct size (8.2±1.2)% vs (38.9±7.9)% were counteracted by ProDTC ( P <0.05 or P <0.01), so was histology. Conclusion\ NF kB might regulate the ischemic myocardial preconditioning.\;
