丁型肝炎病人肝组织中HDAg和肝细胞凋亡双标记表达

Expression of HDAg and hepatocytic apoptosis in liver tissues of patients with hepatitis D

目的 探讨肝细胞凋亡在丁型肝炎发病机制中的作用。方法 采用免疫组化双重染色和TUNEL技术 ,检测 79例丁肝病人肝组织HDAg和肝细胞凋亡分布及其关系 ,以 5 4例乙型肝炎病人肝组织作对照。结果 HDAg以肝细胞核表达为主 ,HDAg、凋亡细胞表达分布与肝组织病理损害程度一致 (P <0 .0 1)。多数凋亡细胞位于HDAg阳性部位或附近 ,肝细胞凋亡在各型肝炎中的分布强度差别有显著性意义(P <0 .0 1)。结论 HDAg表达与肝细胞凋亡分布之间有显著的相关性 。

Objective To study the role of hepatocytic apoptosis in the pathogenesis of hepatitis D.Methods HDAg expression and hepatocytic apoptosis in the samples of liver tissue of 79 patients with hepatitis D were observed with immunohistochemistry and terminal deoxynucleotidyl transferase medited dUTP nick end labeling (TUNEL) respectively. In addition, the relationship of the expression of HDAg with the occurrence of hepatocytic apoptosis in the liver tissue was studied with double labeling and serial sectioning.Results HDAg was expressed mainly in the nucleus of hepatocytes and the degree of expression, extension of HDAg distribution in hepatocytes and hepatocytic apoptosis were obviously correlated to the severity of pathological damages and necrotic and inflammatory reactions of liver tissues(P<0.01). Most of the apoptotic hepatocytes were found in the vicinity of HDAg positive cells. There was a positive correlation of the intensity of hepatocytic apoptosis to HDAg expression(P<0.01).Conclusion Hepatocytic apoptosis may play an important role in the damages of liver cells after HDV infection and there exists close relationship of HDAg expression and hepatocytic apoptosis.
