摘要
目的:探讨C5特异性CD4+T细胞在肺结核患者外周血及在肺病变组织中的分布。方法:应用四聚体检测肺结核患者外周血及肺病变组织C5特异性四聚体阳性CD4+T细胞,同时以健康成人及非结核呼吸道感染患者外周血和肺炎病变组织作为对照。结果:应用C5/HLA-DRB1*090102四聚体检测肺结核组、非结核呼吸道感染组和健康成人组的C5特异性CD4+T细胞的中位数分别为0.15%、0.09%、0.03%,而C5/HLA-DRB1*130201则分别为0.19%、0.10%、0.04%,肺结核患者组外周血C5特异性CD4+T细胞的中位数高于对照组(P﹤0.05)。在肺结核患者的肺病变组织中可观察到C5特异性CD4+T细胞。结论:四聚体技术可用于结核特异性T细胞的检测,为结核特异性T细胞的进一步研究和应用奠定基础。
Objective To detect percentage of peptide C5-specific CD4+ T cells in peripheral bloods (PBLs) and their distribution in the slices of lung tissues from patients with pulmonary tuberculosis (PTB). Methods Two types of tetramers were respectively used to examine C5-specific tetramer-positive CD4~ T cells in the PBLs and pathologic tissues of PTB patients. The PBLs and pathologic tissues of patients with non-TB respiratory infections and healthy adults were used as controls. Results The medians of C5-specific CD4+ T cells from PBLs of patients with pulmonary tuberculosis, patients with pulmonary infection and healthy adults were respectively 0.15%, 0.09% ,and 0.03% by using C5/HLA-DRB1 * 090102 tetramer, while the medians were respectively 0.19%, 0.10%, and 0.04% by using C5/HLA-DRB 1 * 130201 tetramer. The medians of peptide CS-specific CD4+ T cells in PBLs of PTB patients were much higher than in PBLs of control patients (P 〈 0.05). Antigen-specific CD4+T cells were observed in pathologic tissues of PTB patients. Conclusions Tetramer technology can be used to detect tuberculosis-specific CD4+ T cells. It paved a way for further study and application.
