不同表达载体对玉米抗蚜虫基因转化效率的影响 被引量:1

摘要 以玉米自交系87-1和郑22,杂交种87-1×郑22、郑22×87-1、郑58×HiⅡA和昌7-2×HiⅡA的幼胚为外植体,对幼胚的出愈率、愈伤组织的形态差异以及愈伤组织的繁殖继代能力3个方面进行了比较分析.结果表明,昌7-2×HiⅡA产生的愈伤组织是较好的转化受体材料.以农杆菌LBA4404为媒介,将携带有抗蚜虫基因EβF的表达载体pNCX和pBJ40分别转入受体愈伤组织,根据筛选过程中愈伤组织的状态、筛选后所得抗性愈伤组织的块数和最终的抗性苗数,认为以pNCX为载体的基因转化效果明显好于pBJ40载体. The induction frequency, morphological differences, and the subculture ability of the callus from embryo of maize inbred line 87 - 1, Zheng 22, and hybrid 87 - 1 × Zheng 22, Zheng 22 × 87 - 1, Zheng 58 × Hi Ⅱ A, and Chang 7 - 2 × Hi Ⅱ A were analyzed. The results showed that Chang 7 - 2 ×Hi Ⅱ was the best transformed receptor material of these. The expression vectors ( pNCX and pBJ40) carrying anti-aphid gene EβF were transformed to the receptor callus by Agrobacterium tumefa- ciens LBA4404 respectively. It was concluded that transformation efficiency of pNCX was obviously better than one of pBJ40 on the basis of the state of callus, the number of callus pieces and seedlings obtained after resistance screening.
