摘要
目的观察人脐血间充质干细胞(UC-MSCs)静注对单侧输尿管梗阻(UUO)大鼠肾间质纤维化程度的影响。方法将24只大鼠随机分为假手术组、模型组、UC-MSCs组,每组8只。模型组和UC-MSCs组结扎单侧输尿管造成梗阻性肾病模型,UC-MSCs组术后当天即尾静脉注射1×106/mL的UC-MSCs悬液。术后14 d处死各组大鼠,取梗阻侧肾做组织学检查,观察肾间质纤维化的程度;采用免疫组化法检测梗阻侧肾组织中人类白细胞抗原-1(HLA-1)阳性细胞数目和胶原-Ⅳ(COL-Ⅳ)水平。结果假手术组肾组织结构正常。模型组肾小管间质增宽,有较多的炎性细胞浸润和纤维组织增生,肾小管弥漫性扩张,并可见肾小管上皮细胞肿胀、变性、坏死、脱落,小管基底膜不同程度断裂、增厚。UC-MSCs组肾间质纤维化程度明显减轻,肾小管灶状上皮细胞空泡变性,偶见蛋白管型,间质水肿程度减轻。肾间质损伤程度评分假手术组(0.2±0.1)分、模型组(2.1±0.2)分、UC-MSCs组(0.8±0.2)分,两两比较P均<0.05。COL-Ⅳ表达量假手术组0.29±0.06、模型组13.16±8.01、UC-MSCs组7.36±1.07,两两比较P均<0.05。结论 UC-MSCs尾静脉注射可减少肾间质COL-Ⅳ沉积,减轻了肾间质纤维化。
Objective To observe the effect of human umbilical cord mesenchymal stem cells(UC-MSCs) in the renal interstitial fibrosis of unilateral ureteral obstruction (UUO) rats. Methods 24 S-D rats were divided into UC-MSCs group, model group and sham - operated group randomly,8 each. The UC-MSCs group and the model group were established by UUO . The UC-MSCs group cases were injected UC-MSCs 1 × 106/mL to caudal vein of rats after the operation . 14 days later of the operation, all animals were sacrificed. The obstruction kidney was observed in the degree of renal fibrosis through histological examination. The expression of COL-IV and human leauchocyte antigen-1 ( HLA-1 ) in the obstructed kidney was assessed by immunochistochemistry methods. Results The renal tissue structure of Sham operation group was normal. The renal tubule and interstitial of model group was broadening, there were many inflammatory cells infiltration and fibrous tissue hyperplasia, Renal tubular diffuse dilatation, renal tubular epithelial cells were swelling, degenerating, necro- sis and loss,Tubular basement membrane was thickening and fracture of different degree, the UC-MSCs group was obviously decreased. Renal tubular epithelial cells were focal vacuolar degeneration. Protein tube type could be seen occasionally, Interstitial edema was reduced. The score of renal interstitial injury was 0.2 ±0.1 in sham - operated group,2.1 ±0.2 in model group,0. 8 ±0.2 in UC-MSCs group,Comparison of each two groups, P 〈0.05. The expression of COL-IV was 0.29 ± 0.06 in sham - operated group, 13.16 ± 8.01 in model group, 7.36 ± 1.07 in UC-MSCs group, Comparison of each two groups, we all have P 〈 0.05. Conclusion Injection UC-MSCs to UUO rats caudal vein can reduce renal interstitial COL- Ⅳ deposition, remiss renal interstitial fibrosis.
