肝癌组织中HMGA2蛋白、mRNA表达变化及意义 被引量:1

摘要 目的观察肝癌组织中高迁移率族蛋白A2(HMGA2)蛋白、mRNA的表达变化,并探讨其临床意义。方法用免疫组织化学SP法检测30例肝癌及其癌旁组织中的HMGA2蛋白,用RT-PCR法检测HMGA2 mRNA。结果 30例肝癌组织中平均HMGA2阳性率35.1%,而癌旁肝组织HMGA2阳性率<1%,两者HMGA2阳性率相比P<0.01。肝癌组织中HMGA2蛋白表达与肝癌Edmondson分级、肝内转移灶和肝门淋巴结转移有关(P均<0.05),与肿瘤直径无关(P>0.05)。癌旁组织和Ⅰ、Ⅱ、Ⅲ、Ⅳ级肝癌组织中HMGA2 mRNA相对表达量分别为0.03±0.02、0.04±0.06、0.10±0.16、0.52±0.87、0.68±0.95。肝癌组织中HMGA2 mRNA相对表达量明显高于癌旁组织(P均<0.05),肝癌组织中HMGA2 mRNA相对表达量与肝癌Edmondson分级有关(P均<0.05)。结论肝癌组织中HMGA2蛋白、mRNA表达升高,其可能参与肝癌的发生发展过程。 Objective To investigate the significance and their expression of HMGA2 protein and mRNA in hepatocel- lular carcinoma (HCC) and their pericarcinomous tissues (PT). Methods Paraffin embedded tissues from 30 HCC cases and their PT were respectively analysed for HMGA2 protein and mRNA expression by immunohistochemical and RT-PCR method. Results The positive signals were seen in the nucleus. PT of 30 cases were all negative for HMGA2 protein, the positive rate was 0.56%. However, in the HCC 30 cases, 35.1% were positive for HMGA2 protein. The HMGA2 positive rate of HCCs was very significantly higher than that of PT(P 〈 0. 01 ). The relationship between the HMGA2 protein ex- pression in HCC tissues and clinicopathological parameters see tablel. It can be seen from tablel that the HMGA2 protein expression in HCC tissues was related with the Edmondson grade of hepatocellular carcinoma, liver metastases, and liver portal lymph node metastasis ( P 〈 0.05 ) , and was independent of tumor size. The HMGA2 mRNA relative expression in the pericarcinomous tissues and grade I - 1V HCC was respectively O. 03 ±0.02,0.04 ± 0.06,0.10 ± 0.16,0.52 ± 0.87, 0.68 ± 0.95. The HMGA2 mRNA relative expression in HCC was higher than that of the pericarcinomous tissues ( P 〈 0.05 ). The HMGA2 mRNA relative expression in HCC was related with the Edmondson grade(P 〈 0.05 ). Conclusion The HMGA2 protein and mRNA were highly expressed in HCC, which indicated that HMGA2 may be involved in the devel- opment and progression of HCC.
