不同时间点GABA_A受体α_2亚基在神经病理性痛中的表达 被引量:1

Expression of GABA_A Receptor α_2 Subunit in Neuropathic Pain at Different Time Points

摘要 为探讨慢性坐骨神经压榨性损伤(CCI)模型背根神经节GABAA受体α2亚基在不同时间点(3、7、12d)表达。采用雄性SD大鼠28只,随机分为4组(n=7)。A组(假手术组)、B组(CCI术后3d)、C组(CCI术后7d)、D组(CCI术后12d),取L4-L6背根神经节做冰冻切片,用免疫荧光染色方法观察GABAA受体α2亚基表达变化。结果显示,(1)CCI模型鼠热缩足反射潜伏期与假手术组相比显著缩短(P<0.01)。(2)免疫荧光染色示A组、B组、C组、D组DRG神经元均有GABAA受体α2亚基表达,分布于DRG各种直径神经元细胞膜上。(3)与A组比较,D组L4-L6背根神经节GABAA受体α2亚基受体的表达减少(P<0.05);B组、C组变化不明显(P>0.05)。由此可知,慢性坐骨神经压榨性损伤(CCI)模型背根神经节GABAA受体α2亚基表达下调,提示GABAA受体α2亚基参与了神经病理性痛的发生。 To investigate the expression of dorsal root ganglia GABAA receptor α2 subunit in sciatic nerve chronic crush injury (CCI) model at different time points (3,7,12 d). Methods:Twenty-eight male Sprague Dawley rats were randomly divided into 4 groups:group A of sham operation ,group t3 3 days after CCI;group C 7 days after CCI,and group D 12 days after CCI. L4-L6 dorsal root ganglia were used for frozen sections. Immunofluorescence staining was applied to observe the changes in GABAA receptor α2 subunit. Results : (1) CCI injured side produced significantly thermal with drawal latency shorter ( P〈 0.01 ). (2) The immunofluorescence staining show A,B,C and D group of DRG neuron nave GABAA receptor α2 subunit expression,located in the DRG each kind of diameter neuron cell membrane. (3)Compared with group A, the expression of L4-L6 dorsal root ganglia GABAA receptor variousaz subunit receptor in group D decreased (P〈0.05) ;no significant changes in group 13 and group C (P 〉0.05). Conclusion:The decrease in the expression of GABAA receptor α2 subunit in sciatic nerve chronic crush injury (CCI) model of dorsal root ganglia suggests that GABAA receptor α2 subunit has a correlation with neuropathic pain.
