摘要
采用油茶上胚轴、下胚轴和子叶作外植体进行油茶组织离体培养诱导愈伤组织发生和不定芽分化的实验。实验结果表明,0.2%的HgCl2消毒处理油茶种胚外植体10 min效果较好;油茶上胚轴、下胚轴和子叶组织在MS+2,4-D 2 mg/L+KT0.5 mg/L的培养基上,愈伤组织诱导率分别为95.0%、66.7%和30.0%;在MS+6-BA2.5 mg/L+KT 0.1 mg/L+NAA 0.5 mg/L培养基上,不定芽诱导率分别是66.7%、33.3%和11.1%;以MS+6-BA 2 mg/L+NAA 0.2 mg/L继代培养,上胚轴、下胚轴和子叶所形成愈伤组织芽增殖系数40天时为5.8、5.6和5.3。
Taking the epicotyls, hypocotyls and cotyledons of camellia as the materials, the callus induction effects and differentiation of adventitious bud were studied. Using the improved MS as the basic medium, sterilizing the ma- terials with HgC12 of 0. 2 %, the callus induction ratio of the epicotyls, hypocotyls and cotyledons on MS + 2, 4 - D 2mg/L + KT O. 5 mg,/L were 95.0 % , 66.7 % and 30. 0 % respectively, the induction ratio of adventitious bud on MS + 6-BA2.5 mg/L + KT0. lmg/L + NAA0.5mg/Lwere66.7 %, 33.3 % and 11.1% respectively. U- sing the formulation of MS + 6 - BA 2 mg/L + NAA 0.2 mg/L for subculture, 40 days after treatment, the bud proliferation coefficients of the epicotyls, hypocotyls and cotyledons were 5.8, 5.6 and 5.3 respectively.
