摘要 目的观察鼻咽癌组织中磷脂酰肌醇3-激酶(phosphatidylinositol 3-kinase PI3K)的表达与临床病理的关系,探讨其可能参与的鼻咽癌的侵袭转移过程的机制。方法采用免疫组化S-P法及Q-RT-PCR分别在蛋白质与核酸水平检测鼻咽癌标本肿瘤组织与鼻咽正常上皮组织中PI3K的表达。并探讨其表达与临床指标的关系,及其在鼻咽癌侵袭转移中的作用。结果 (1)鼻咽正常上皮组织PI3K的表达显著低于鼻咽癌组,两者差异有统计学意义(P<0.05)。(2)PI3K的表达与患者的年龄、性别以及肿瘤原发部位和大小均无关(P>0.05),而与分化程度、浸润深度和TNM分期等临床病理指标之间的差异比较则显示统计学意义显著(P<0.05)。结论 PI3K在鼻咽癌的恶性表型及侵袭和转移中起促进作用,它的表达同TNM分期呈正向关。 Objective the aim of this study is to identify whether the overexpression of PI3K is inter-related in NPC, and to define their association with the extent of the presenting disease as measured by the TNM staging system. Methods we measured the expression of PI3K in NPC at the genetic,transcriptional,and translational level. NPC samples (n=102) were analyzed with immunohistochemical labeling for PI3K protein, twenty eight of which were also analyzed with quantitative real-time polymerase chain reaction (Q-RT-PCR) and western blots for PI3K mRNA and protein expression. The expression of P13K in normal nasopharyngeal tissue (n=26) acted as control. The expression was gauged with regard to stage,grade. Results PI3K was overexpressed in NPC cells when compared with normal nasopharyngeal tissue and their protein up-regulation was significantly associated with tumor-nodemetastasis (TNM) stage (P〈0.05). Conclusion Our findings suggest that PI3K overexpression is common abnormalities in NPC. It is also in- dicated that their up-regulation is correlated with tumor progression and metastatic phenotype.
