摘要
为了得到满足分子生物学要求的高质量大花蕙兰子房DNA,以大花蕙兰子房为材料,采用SDS法、CTAB法、优化CTAB法、高盐CTAB法和试剂盒法提取子房基因组DNA。通过琼脂糖凝胶电泳和仪器检测提取产物,结果表明,采用优化CTAB法提取的DNA产物浓度高,纯度高,无酚类、多糖等杂质的污染,证明优化CTAB法是提取大花蕙兰子房基因组DNA的最适方法。
In order to get the genomic DNA of Cymbidium's ovary with high quality, which can meet the requirements of molecular biology researches, the Cymbidium's ovary was selected as the materials to extract its genomic DNA by five different methods (SDS, CTAB, optimized CTAB, high-salt CTAB and Kit method) . The extraction products were detected by gel electrophoresis and instrument. The results indicate that the extraction products from optimized CTAB were of high concentration and high purity without phenols, polysaccharides and other impurities. Optimized CTAB was the optimum method for extracting the genomic DNA of Cymbidium's ovary.
