大肠癌ColⅣ、LN及FN的免疫组化研究 被引量:1

Expression of Laminin,Fibronectin and Collagen Ⅳ in Colorectal Carcinomas

摘要 目的 :检测基底膜主要成分层粘连蛋白 (LN)、纤维粘结蛋白 (FN)及Ⅳ型胶原纤维 (ColⅣ )在大肠癌中的表达以及与浸润、转移的关系。方法 :免疫组化标记大肠癌组织中ColⅣ、LN及FN。结果 :大肠癌组织中基底膜成分存在不同程度的缺失 ,表现为ColⅣ、LN呈不连续、片状分布 ;癌组织分化越低 ,LN缺失越明显 ;ColⅣ表达与组织类型有显著关系 ,而与分化程度无关。一般腺癌阳性率明显高于粘液腺癌及印戒细胞癌 ;DukesB、C期大肠癌ColⅣ表达阳性率低于A期大肠癌 ;FN表达与上述因素关系不大。结论 :大肠癌基底膜成分的缺失与其侵袭、转移能力有关 ,尤其是ColⅣ的缺失可能标志其具较强的侵袭、转移能力。 Purpose: This study sought to determine if there is a correlation between the expression of major components of basement membrane such as collagen Ⅳ , fibronectin, laminin etc. and the invasion and metastasis of colorectal carcinomas. Methods: An immunohistochemical study of the major basement membrane antigen collagen Ⅳ , fibronectin and laminin was performed and 109 cases of colorectal carcinoma were observed. Results: There was a statistically significant correlation between the collagen Ⅳ expression and histologic type of colorectal carcinoma. The depletion of collagen Ⅳ was more prominent in mucinous carcinoma and signet carcinoma than that in adenocarcinoma, but no close correlation was found between the expression of collagen Ⅳ and the histologic stage of tumor. Only about 30% of the stage B and stage C tumors expressed collagen Ⅳ . This percentage was slightly lower than that of stage A tumors. In addition, laminin or fibronectin expression did not show any significant association with these parameters mentioned above. Conclusion: The absence of the components of basement membrane especially the collagen Ⅳ is intimately associated with invasion and metastasis in the case of colorectal carcinomas.
