药物H、Y、D对大鼠肝脏低温保存损伤的保护作用

摘要 目的 研究药物H、Y、D对大鼠肝脏低温保存损伤的保护作用。方法 采用大鼠肝脏离体非循环灌注模型 ,观察乳酸林格液 (LR液 )中加入不同剂量药物H、Y、D后保存大鼠肝脏 12h的效果。结果 鼠肝经加入药物H、Y、D的LR液低温保存后 ,其肝组织ATP、TAN含量、AEC及分泌胆汁量明显高于对照组 (P <0 0 5 ) ,且ATP、分泌胆汁量与LR液内H、Y、D剂量在一定范围内呈正相关。药物H、Y、D具有抗·OH的作用 ,可减轻对肝细胞损伤。结论 药物H、Y、D可明显提高供体肝脏的保护效果 。 Objective To research the protective effects of drugs H,Y and D on cold preservation injury of rat liver.Methods We compared the preservation effect of Lactate Ringer's(LR) solution including drugs H,Y and D with that of LR solution added nothing by using isolated nonrecirculated perfusion rat liver model. Results After the rat's livers were preserved for 12 hours in LR solution with the addition of drugs H,Y and D,the contents of ATP,TAN,AEC and bile production were significantly higher than that of the control group( P <0.05).Within certain range of the dosage of drugs H,Y and D,the contents of ATP and bile production were in the positive ration with the dosage in the LR solution.We also found that drugs H,Y and D had a marked protection against hydroxyl free radical and alleviated the injury on liver cell.Conclusion The significant protection of drugs H,Y and D on liver preservation revealed in this research may act by a mechanism which correlated to the effect of improving the energy metabolism of cold storage liver and resisting free radical.
