摘要
通过紫外诱变提高嗜酸乳杆菌的产乳酸能力,通过正交试验确定了诱变参数:20 W紫外灯,菌液稀释至OD6000.65左右,照射60 s,照射距离为30.5 cm,此条件的致死率为92.5%。诱变处理后的菌体产酸速率明显高于出发菌株,突变菌株在培养36 h时产酸量达到最大值10.83 g/L,而出发菌株最大值为10.41 g/L,且突变菌株具有稳定的遗传性。
The study was designed to enhance the ability of Screening of high lactic acid-producing Lactobacillus acidophihts by UV mutation. Parameters with a lethal rate 92.5% for UV mutation determined by orthogonal test were 20W UV,bacteria was diluted to around OD60o 0.65,illuminate at a distance of 30.5cm for 60s. The mutant was significantly higher than the original strains. The mutant produced 10.83 g / L lactic acid when 36 hours and detected no increase later. The original strains produced lactic acid maximum of 10.41 g / L and the mutant strain keeps good hereditary stability.
