S-腺苷蛋氨酸对大鼠肝缺血再灌注损伤后清蛋白mRNA表达的影响 被引量:5

Influence of S-ademetionine on albumin mRNA expression in rat liver after ischemia-reperfusion injury

摘要 目的:探讨S-腺苷蛋氨酸(思美泰)对大鼠肝缺血再灌注损伤后清蛋白(ALB)mRNA表达的影响。方法:将72只SD大鼠随机均分为假手术组,肝缺血再灌注组(IR组),肝缺血再灌注+腺苷蛋氨酸干预组(SAM组)。IR组和SAM组均以肝脏缺血20 min后再灌注的方式造模。SAM组于术后1 h腹腔注射思美泰100 mg/(kg.d),假手术组和IR组予以等体积生理盐水。各组分别于术后1,4,7,10 d处死6只大鼠,用实时定量PCR方法测定肝组织ALB mRNA的含量。结果:术后第1天SAM组和IR组ALB mRNA相对表达量较假手术组明显降低(均P<0.05),但两组间差异无统计学意义(P>0.05);从术后第4天开始,SAM组ALB mRNA的相对表达量明显高于IR组(P<0.05),且至术后第10天明显超过假手术组(P<0.05)。结论:腺苷蛋氨酸能提高肝缺血再灌注损伤后ALB mRNA的合成,增强肝脏的合成功能。 Objective: To investigate the influence of S-ademetionine on albumin(ALB) mRNA expression in rat liver after ischemia-reperfusion injury. Methods: Seventy-two male SD rats were equally randomized into sham operation group,hepatic ischemia-reperfusion group(IR group),and hepatic ischemia-reperfusion plus S-ademetionine treatment group(SAM group).The animal models were induced by 20 min hepatic ischemia and subsequent reperfusion in IR group and SAM group.One hour after surgery,rats of SAM group were given S-ademetionine [100 mg/(kg?d)] by intraperitoneal injection,and rats of sham operation group and IR group were given normal saline at the same volume instead.Six rats from each group were sacrificed on postoperative day(POD) 1,4,7 and 10,respectively,and the ALB mRNA expressions in the liver tissues were detected by quantitative real-time RT-PCR. Results: On POD 1,the relative expression levels of ALB mRNA in SAM group and IR group were significantly lower than that of sham operation group(both P0.05),but no significant difference was noted between themselves(P0.05).The relative expression levels of ALB mRNA of SAM group began to increase from POD 4 compared with IR group(all P0.05),and were even higher than that of sham operation group on POD 10(P0.05). Conclusion: S-ademethionine can increase ALB mRNA expression in rat liver after ischemia-reperfusion injury,and boost liver synthetical function.
