摘要
通过比较水稻野生型(WT)和光敏色素B突变体(phyB)的基因表达图谱,筛选出一个受phyB调控、编码植物特有串联锌指(TZF)蛋白的基因OsDOS,并分析了该基因编码蛋白及其启动区的生物信息学特征,以及该基因的表达模式。结果显示,OsDOS基因在phyB突变体中的表达水平明显高于野生型,推测phyB感受的光信号抑制OsDOS基因的表达。OsDOS基因在叶片中表达最高,其次是萌发的胚。另外,通过检测NaCl和PEG处理后WT水稻中OsDOS基因的表达情况,发现盐和干旱胁迫可能抑制该基因的表达;在ABA处理后OsDOS基因的表达模式与上述非生物胁迫处理的结果相似,因此推测NaCl和PEG非生物胁迫可能通过ABA途径而影响OsDOS基因的表达。本研究结果为深入分析OsDOS基因在水稻生长发育中的作用奠定了基础。
One plant-unique TZF (tandem zinc finger) gene, OsDOS, regulated by phyB was identi- fied by comparing the gene profiles between wild type (WT) and phytochrome B mutant (phyB) of flee. The bioinformation characteristics of OsDOS protein and promoter region of OsDOS were analyzed, as well as its ex- pression patterns. The results showed that OsDOS gene expressed higher in phyB mutant than in WT, sugges- ting that phyB - mediated light signal repressed the expression of OsDOS. Organ specificity of its expression re- vealed that OsDOS gene expressed relatively higher in leaves, and then in germinating embryos. Moreover, the expression level of OsDOS was reduced by NaC1, PEG and ABA. Thus, it was speculated that the effect of NaC1 and PEG on OsDOS gene expression might be could lay foundation for analyzing the roles of OsDOS regulated through the ABA pathway in rice. This study gene in rice development.
