抗疏健骨颗粒对骨质疏松模型大鼠骨密度和骨生物力学的影响 被引量:6

摘要 目的:观察抗疏健骨颗粒对骨质疏松模型大鼠骨密度和骨生物力学的影响,研究其防治骨质疏松症的作用机理。方法:60只大鼠喂养1周后随机分为假手术组(10只)和去势组(50只)。去势组随机分5组,模型组,尼尔雌醇组,抗疏健骨颗粒小、中、大剂量组,每组10只,除假手术组外,其余5组大鼠行卵巢切除术造模。抗疏健骨颗粒大、中、小剂量组分别给予抗疏健骨颗粒混悬液0.4 g/mL、0.2 g/mL、0.1 g/mL,2 mL/d,每日1次,尼尔雌醇组给予尼尔雌醇片(1 mg/kg体质量),配成0.15 g/L混悬液,1周1次,每次2 mL,模型组和假手术组以生理盐水(8 mL/kg体质量)灌胃。12周后测定离体骨密度。计算最大载荷、断裂挠度、弯曲能量。结果:与模型组比较,抗疏健骨颗粒大、中剂量组的腰椎骨和股骨远端骨密度均显著增加(P<0.01),抗疏健骨颗粒大、中剂量组与尼尔雌醇组的最大载荷显著增高(P<0.01),但小剂量组增加不明显(P>0.05);抗疏健骨颗粒各剂量组断裂挠度与模型组比较,虽有升高趋势,但变化不明显(P>0.05);尼尔雌醇组、抗疏健骨颗粒大剂量组的弯曲能量与模型组比较,增高显著(P<0.01),中、小剂量组虽有升高趋势,但无显著差异(P>0.05)。结论:抗疏健骨颗粒能够改善骨密度和骨生物力学性能,恢复骨代谢平衡,从而达到防治骨质疏松的作用。 Objective: To observe the influence of Kangshu Jiangu Granules on bone density and femoral biomechanics of the osteo- porotic model rats and study the mechanism of preventing and treating osteoporosis. Methods : 60 rats fed after one week were ran- domly divided into sham operation group (10) and castration group (50). Castration group were randomly divided into five smaller groups with ten rats in each group: model group, neal estradiol group, small, medium and large dose groups of Kangshu Jiangu Granules. Except the sham operation group, other 5 groups were made models with oophorectomy. The small, medium and large dose groups of Kangshu Jiangu Granules were respectively given suspension liquid of Kangshu Jiangu Granules 0. 4 g/mL, 0.2 g/mL, 0. 1 g/mL, 2 mL/d once a day. Neal estradiol group were given 0. 15 mg/mL suspension liquid of Neal estradiol ( 1 mg/kg weight) once a week and 2 mL each time. Model group and sham operation group were given gastrol perfuson of physio- logical saline (8 mL/kg weight) . 12 weeks later, the isolated bone density was measured to calculate the maximum load calcula- tion, the maximum deflection and bending energy. Results : The bone density of the lumbar vertebrae and the disral end of the fe- mur of the small, medium and large dose groups of Kangshu Jiangn Granules were significantly increased ( P 〈 0.01 ) compared with those of the model group ; the maximum load in the group of the small, medium and large dose groups of Kangshu Jiangu Granules and neal estradiol group increased significantly ( P 〈 0.01 ), but the increase in the group of small dose is not obvious ( P 〉 0.05 ) ; The crack deflection of all groups of Kangshu Jiangu Granules had an unobvious rise trend (P 〉 0.05 ) compared with the model group; the bending energies of neal estradiol group and the large dose groups of Kangshu Jiangu Granules increased significantly (P 〈0.01 )compared with that of the model group; the medium and small dose group had an increase trend, but there was no sig- nificant difference (P 〉0.05). Conclusion: Kangshu Jiangu Granules can improve the bone density and bone biomechanical per- formance, restore bone metabolism balance, so as to achieve the function of preventing osteoporosis.
