抗弧菌海洋细菌BN-1菌株几种胞外酶活性的测定 被引量:2

摘要 采用平板透明圈法初步测定海洋细菌BN-1菌株产淀粉酶、蛋白酶、脂肪酶和纤维素酶活性。试验结果表明,BN-1菌在淀粉酶和蛋白酶鉴别培养基上能形成明显清晰的透明圈,具有较强的产蛋白酶、淀粉酶能力;在脂肪、纤维素鉴别培养基上形成的透明圈直径较小。比色法测定结果表明,BN-1菌株产淀粉酶和蛋白酶能力较强,培养16h蛋白酶活性高达1076.14U/mL,培养18h淀粉酶活性可达1053.49U/mL。培养16h纤维素酶活性高达127.03U/mL,20h脂肪酶活性可达474.83U/mL。 The activities of amylase,protease,lipase and cellulase in marine bacterium BN-1 were detected using plate culture and clear zones methods.The results showed that the clear zones formed on the amylase selective plate and protease selective plate,and the strain had relatively strong activities to produce amylase and protease.However,the strain was found to produce smaller zone diameters on lipase selective plate and cellulase selective plate.Then,detection of the four enzyme activities by colorimetric method revealed that the activities of amylase and protease were found stronger than the activities of lipase and cellulase.The maximal activity(1076.14 U/mL) of amylase was observed 16 h after culture,and the maximal activity(1053.49 U/mL) of protease was found 18 h after culture.However,there was the activity(127.03 U/mL) of cellulase,as the minimal value,16 h after culture and the activity of lipase(474.83 U/mL) 20 h after culture.
