摘要
目的探讨血清脂联素蛋白(APN)及肝脏脂联素受体2 mRNA的表达在实验性大鼠肝纤维化形成和发展中的作用。方法将SD大鼠随机分为对照组与模型组,两组大鼠腹部皮下分别注射100%橄榄油和40%CCl4-橄榄油溶液,每周2次,共10周,于第2、6、10周末分批取材,分别抽取各大鼠的血液及切取各实验大鼠的肝脏标本,以ELISA法测定大鼠APN蛋白表达情况,RT-PCR法检测肝中脂联素受体2 mRNA的表达,常规石蜡切片HE染色观察各实验大鼠肝脏形态学改变。结果模型组大鼠APN水平随造模时间延长逐步降低,同时脂联素受体2 mRNA表达同步下降,各时间点模型组与正常对照组比较差异有统计学意义(P<0.05)。结论 APN水平及肝脏脂联素受体表达异常可能在肝纤维化的形成和发展中起一定作用。
Objective To study the significance of expression of serum adiponectin and liver adiponectin receptor 2 mRNA in development of liver fibrosis in experimental rats.Methods SD rats were randomly divided into control group and model group,rats in model group were injected with 100% olive oil and 40% carbon tetrachloride solusion twice every week for 10 weeks.Samples of rat sera and liver tissues were obtained at the end of 2nd,6th,and 10th week.The expression of serum adiponectin was evaluated by ELISA.The expression of hepatic adiponectin receptor 2 mRNA was measured with realtime-polymerase chain reaction.Sections of liver tissue with HE staining were examined for pathological changes.Results In model group,levels of serum adiponectin were gradually decreased,meanwhile,the expression of adiponectin receptor 2 mRNA appeared to be falling down.Significant differences were observed at each time point between rats in model and control groups(P0.05).Conclusion The variation in expression of adiponectin and its receptor 2 mRNA may be critical in pathogenesis and development of liver fibrosis.
