摘要
通过比较了单层蔗糖分步离心和不连续蔗糖梯度离心制备的大鼠脑突触体对5-羟色胺和去甲肾上腺素的再摄取能力。结果表明,采用分步离心法可减少制备突触体的时间,降低操作难度,所得脑突触体具有有效蛋白含量较高、稳定性较好的优势。该法所制备的突触体对相应递质的特异性摄取能力有所提高。
The 5-hydroxytryptamine and noradrenalin reuptake capabilities of rat brain synaptosome prepared by step-by-step centrifugation with single-layer sucrose and discontinuous sucrose gradient centrifugation were compared. The results showed that the step-by-step centrifugation method could reduce the preparation time and reduce the operation difficulty, and the obtained brain synaptosome had the advantages of a higher effective protein content and better stability. The uptake capability of specific transmitters by the prepared synaptosomes was improved.
