摘要
糖基化作为蛋白质翻译后最重要的修饰之一,对于蛋白质的折叠、运输和定位起着重要作用,与重组糖蛋白在体内的生物活性、半衰期和分解代谢等密切相关。蛋白质的糖基化过程受很多因素的影响,包括细胞种类、培养基的差异、培养过程的不同控制、高尔基体的转运时间、细胞的状态及细胞内核酸等前体物质的利用等。通过优化条件提高蛋白质的糖基化,保证不同批次间蛋白质糖基化的一致性,将会大大增加重组糖蛋白药物的治疗效果。本文从不同方面论述了蛋白质糖基化的方法。
As one of the most important posttranslational modifications, glycosylation is crucial for the refolding, transportation and location of proteins. It is also related with their bioactivities, metabolic circulation and degradation in vivo. The glycosylation process was affected by many factors including cells, mediums and culture process, the transit time in the Golgi, cell status and the availability of intracellular nucleic acid precursors. Therapeutic efficacy will be improved by optimization of glycosylation levels and assurance of the consistent glycosylation. This paper discusses the methods of improving glycosylation of glycoprotein medicines, which will be very useful for the recombinant glycoprotein production.
