摘要
目的比较研究乙型肝炎病毒(HBV)转基因小鼠和C57BL/6小鼠骨髓间充质干细胞(bone marrowmesenchymal stem cells,BMSCs)的分离培养体系。方法采用贴壁培养法建立HBV转基因小鼠和C57BL/6小鼠BMSCs分离培养体系,通过在培养液中添加不同浓度的血清进行培养,分析HBV转基因小鼠和C57BL/6小鼠BMSCs细胞形态和数量的差异。结果 C57BL/6小鼠原代BMSCs生长迅速,按1∶3传代后呈漩涡状生长,形态类似成纤维细胞。HBV转基因小鼠原代BMSCs生长缓慢;以1∶2传代后生长缓慢,有明显的血清依赖性及密度依赖性。结论在相同的生长环境下,不同血清浓度的培养液中,HBV转基因小鼠BMSCs生长较正常C57BL/6小鼠的BMSCs生长明显缓慢。
Objective To carry on a comparative study of the isolation and culture system of HBV transgen- ic mice and C57BL/6 mice bone marrow mesenchymal stem cells (BMSCs). Methods HBV transgenic mice and C57BL/6 mice BMSCs were isolated and cultured by adherent culture method. Different concen trations of serum was added to culture medium. Analyzed HBV transgenic mice and C57BL/6 mouse BM-SCs cell morphology and number of differences. Results C57BL/6 mice primary BMSCs grew rapidly, then it was swirling growth according to the 1 : 3 passage, which of morphology similar to fibroblasts. HBV transgenic mice primary BMSCs grew slowly, according to 1 : 2 passage, which of growth was slow, and had a strong dependence of serum and cell density. Conclusion Compared with C57BL/6 mice, HBV transgenic mice BMSCs exist in serum-dependent cell growth and density dependence in same growth con- dition in vitro .
