氧化锌对猪肠上皮细胞HSP70 mRNA表达和IL-10分泌的影响 被引量:4

摘要 本试验旨在研究氧化锌与猪肠上皮细胞抗应激和抗炎症能力的关系。试验以体外培养的猪肠上皮细胞系(IPEC-J2细胞系)为模型,考察在有、无脂多糖(LPS)条件下,不同水平氧化锌(0、0.05、0.10 mmol/L和0.15 mmol/L)在不同培养时间0.5、1.5、4.5 h和24 h对细胞HSP70 mR NA表达和IL-10分泌的影响。结果表明:猪肠上皮细胞与氧化锌共培养较长(1.5 h和4.5 h)和长时间(24 h),HSP70 mR NA表达随氧化锌剂量的增加呈线性增加(P<0.01),且高剂量氧化锌(0.10 mmol/L和0.15 mmol/L)显著促进HSP70 mR NA表达(P<0.05),并维持其长期稳定性。高剂量短时间和低剂量长时间的氧化锌处理,显著提高细胞培养液中IL-10水平(P<0.05)。表明高剂量氧化锌可通过促进HSP70基因表达和IL-10分泌,提高猪肠上皮细胞抗应激和抗炎能力。
